A 3D Cell‐Free Bone Model Shows Collagen Mineralization is Driven and Controlled by the Matrix

چکیده
Osteons, the main organizational components of human compact bone, are cylindrical structures composed layers mineralized collagen fibrils, called lamellae. These lamellae have different orientations, degrees organization, and mineralization where intrafibrillar extrafibrillar minerals intergrown into one continuous network oriented crystals. While cellular activity is clearly source organic matrix, recent in vitro studies call question whether cells also involved matrix suggest that this process could be simply driven by interactions mineral with extracellular matrix. Through remineralization demineralized bone complete multiscale reconstruction 3D structure composition osteon without involvement demonstrated. Then, cell-free system explored as a realistic, functional model for situ investigation matrix-controlled processes. Combined Raman electron microscopy indicate glycosaminoglycans (GAGs) play more prominent role than generally assumed matrix–mineral interactions. The experiments show organization part result its interaction developing mineral.

Passaged primary cultures of young adult rat bone marrow cells were maintained in medium containing combinations of the supplements dexamethasone, ascorbic acid and Na-{3-glycerophosphate. The cultures were examined by both light and scanning electron microscopy (with additional energy dispersive X-ray analysis). In cultures with fully supplemented medium, an extracellular matrix formed at the ...
متن کاملa study on insurer solvency by panel data model: the case of iranian insurance market
the aim of this thesis is an approach for assessing insurer’s solvency for iranian insurance companies. we use of economic data with both time series and cross-sectional variation, thus by using the panel data model will survey the insurer solvency.
Matrix vesicle-mediated mineralization in bone
Matrix vesicle-mediated mineralization is orchestrated by ultrastructural and biochemical events that lead to crystal nucleation and growth. Osteoblasts secrete extracellular matrix vesicles equipped with a variety of membrane transporters and enzymes, which are necessary for the initial nucleation and subsequent growth of calcium phosphate crystals. The influx of phosphate ions into the matrix...
متن کاملMathematical model for bone mineralization
Defective bone mineralization has serious clinical manifestations, including deformities and fractures, but the regulation of this extracellular process is not fully understood. We have developed a mathematical model consisting of ordinary differential equations that describe collagen maturation, production and degradation of inhibitors, and mineral nucleation and growth. We examined the roles ...
متن کاملMathematical model for the mineralization of bone.
A mathematical model is presented for the transport and precipitation of mineral in refilling osteons. One goal of this model was to explain calcification "halos," in which the bone near the haversian canal is more highly mineralized than the more peripheral lamellae, which have been mineralizing longer. It was assumed that the precipitation rate of mineral is proportional to the difference bet...
